碩士 / 國立清華大學 / 分子與細胞生物研究所 / 101 / 長久以來魚類標本的製作多著重於抗菌防腐以達型態維持,且魚體內存在著多 種色素,各色素之性質不同,最適保存條件亦不相同。因此對於體色保存一直沒 有多大突破,標本只能靠人工上色以期能表現出原體色。 本實驗採用之黃唇色鯛(Labidochromis caeruleus)具有鮮黃的體色,其色素主要 成分為類胡蘿蔔素(carotenoid)。Carotenoid 十分不穩定,容易因溫度、光照、酸 鹼與氧化壓力而降解,為測詴適合保存carotenoid 之環境與保存液,採用了多種 照度、溫度、酸鹼梯度及抗氧化劑並以色度計檢測其保存能力,結果發現抗壞血 酸(Ascorbic acid: AA)配合亞硫酸氫鈉(Sodium bisulfate: SB)兩類抗氧化劑可得最 佳保存效果,並在強光與高溫下穩定。進行Carotenoid 萃取分析亦發現含量隨時 間只有少許下降,但色澤並無太大改變,已十分優於傳統配方。 結果顯示本配方可做為有效保存黃唇色鯛標本的方式,亦有進一步做為其他魚 種保存的可能。
